Deuteration of nonexchangeable protons on proteins affects their thermal stability, side‐chain dynamics, and hydrophobicity

Published: 26 May 2020

Author(s): Parker J. Nichols, Isaac Falconer, Aaron Griffin, Colin Mant, Robert Hodges, Christopher J. McKnight, Beat Vögeli, Liliya Vugmeyster

Abstract We have investigated the effect of deuteration of non‐exchangeable protons on protein global thermal stability, hydrophobicity, and local flexibility using well‐known thermostable model systems such as the villin headpiece subdomain (HP36) and the third immunoglobulin G‐binding domain of protein G (GB3). Reversed‐phase high‐performance liquid chromatography (RP‐HPLC) measurements as a function of temperature probe global thermal stability in the presence of acetonitrile, while...

Structural and chemical trapping of flavin‐oxide intermediates reveals substrate‐directed reaction multiplicity

Published: 26 May 2020

Author(s): Kuan‐Hung Lin, Syue‐Yi Lyu, Hsien‐Wei Yeh, Yi‐Shan Li, Ning‐Shian Hsu, Chun‐Man Huang, Yung‐Lin Wang, Hao‐Wei Shih, Zhe‐Chong Wang, Chang‐Jer Wu, Tsung‐Lin Li

Abstract Though reactive flavin‐N5/C4α‐oxide intermediates can be spectroscopically profiled for some flavin‐assisted enzymatic reactions, their exact chemical configurations are hardly visualized. Structural systems biology and stable isotopic labelling techniques were exploited to correct this stereotypical view. Three transition‐like complexes, the α‐ketoacid…N5‐FMNox complex (I), the FMNox‐N5‐aloxyl‐C′α−‐C4α+ zwitterion (II), and the FMN‐N5‐ethenol‐N5‐C4α‐epoxide (III), were determined...

Activation mechanism of plasmepsins, pepsin‐like aspartic proteases from Plasmodium, follows a unique trans‐activation pathway

Published: 26 May 2020

Author(s): Ishan Rathore, Vandana Mishra, Chandan Patel, Huogen Xiao, Alla Gustchina, Alexander Wlodawer, Rickey Y. Yada, Prasenjit Bhaumik

Plasmodium parasites that cause malaria produce plasmepsins (PMs), pepsin‐like aspartic proteases that are important antimalarial drug targets due to their role in host hemoglobin degradation. The enzymes are synthesized as inactive zymogens (pro‐PMs), and the mechanism of their conversion to the active, mature forms has not been clearly elucidated. Our structural investigations of vacuolar pro‐PMs with truncated prosegment (pro‐tPMs) reveal that the formation of the S‐shaped dimer is their...

A New Robust Epigenetic Model for Forensic Age Prediction

Published: 26 May 2020

Author(s): Alberto Montesanto, Patrizia D’Aquila, Vincenzo Lagani, Ersilia Paparazzo, Silvana Geracitano, Laura Formentini, Robertina Giacconi, Maurizio Cardelli, Mauro Provinciali, Dina Bellizzi, Giuseppe Passarino

Abstract Forensic DNA phenotyping refers to an emerging field of forensic sciences aimed at the prediction of externally visible characteristics of unknown sample donors directly from biological materials. The aging process significantly affects most of the above characteristics making the development of a reliable method of age prediction very important. Today, the so‐called “epigenetic clocks” represent the most accurate models for age prediction. Since they are technically not achievable in...

Temperature‐Induced Structure Reconstruction to Prepare a Thermally Stable Single‐Atom Platinum Catalyst

Published: 26 May 2020

Author(s): Dongxu Yan, Jing Chen, Hongpeng Jia

Abstract Single‐atom noble metals on a catalyst support tend to migrate and agglomerate into nanoparticles owing to high surface free energy at elevated temperatures. Temperature‐induced structure reconstruction of a support can firmly anchor single‐atom Pt species to adapt to a high‐temperature environment. We used Mn3O4 as a restructurable support to load single‐atom Pt and further turned into single‐atom Pt‐on‐Mn2O3 catalyst via high‐temperature treatment, which is extremely stable under...

Effect of Pre‐Strain Aging on the Microdamage Properties of Composite Solid Propellant

Published: 26 May 2020

Author(s): Dong‐Mo Zhou, Han‐Xu Wu, Xiang‐Yang Liu, Peng‐Jun Zhang, Hui‐Yuan Wang

Abstract Accelerated aging tests under pre‐strain were conducted on HTPB‐based composite solid propellant with the goal of investigating the effect of pre‐strain aging on its microdamage properties. The tensile fracture morphologies, stress‐strain curve and dissipative energy density of propellant samples were analyzed. Results showed that there was no obvious dewetting macroscopically when the pre‐strain was less than 9 %, but the pre‐strain can still cause microdamage of propellant...

Effects of Superimposed Pressure on the Mechanical Properties of HTPB Propellant in a Wide Temperature Range

Published: 26 May 2020

Author(s): Liang Zhang, Zhi‐bin Shen, Hai‐yang Li

Abstract Considering the propellant of solid rocket motor with tube grain is in the state of superimposed pressure after ignition, the effect of pressures, temperatures, and tensile rates on the mechanical properties of solid propellant were investigated together. A test system with wide temperature‐ambient pressure range was developed to investigate the effects of superimposed pressure on the performance of propellant. The uniaxial constant tensile rate tests for a certain HTPB propellants...

Method to Determine the Impact Strength of Triple‐Base Gun Propellant Based on Strain Energy Density

Published: 26 May 2020

Author(s): Ji‐Hua Liu, Hong‐Li Zhao, Chang‐Hui He, Jian‐Wei Jin, Bao‐Ming Zhao, Qiong‐Lin Wang, Zou‐Zou Zhang

Abstract A method to obtain the impact strength of triple‐base gun propellant is proposed according to strain energy density. Using a drop‐weight impact testing machine, the impact strengths of DAGR125‐21/19 gun propellant at room temperature (20 °C) and low temperature (−40 °C) were studied. In the experiments, there were different impact energies via changing the drop weight or height. The surface images of gun propellants during the impact process were captured using a high‐speed camera....

Preparation, Thermal and Sensitivity Properties of Nano‐Sized Spherical Nitrocellulose Composite Crystal

Published: 26 May 2020

Author(s): Xiangjun Meng, Chengkai Pu, Pengteng Cui, Zhenggang Xiao

Abstract The sensitivity study of propellants has been the concern of many researchers, and many achievements have been gained in this field. Nitrocellulose was replaced by some energetic thermoplastic elastomers to prepare insensitive high energy propellants, which is an effective method for reducing the sensitivity of propellants. However, the propellants based on these binders still have some defects. Preparation and application of insensitive nitrocellulose may overcome these drawbacks and...
