Searching for analgesic drug candidates alleviating oxaliplatin‐induced cold hypersensitivity in mice

Published: 22 March 2019

Author(s): Kinga Sałat, Anna Furgała, Natalia Malikowska‐Racia

Abstract Oxaliplatin is a third‐generation, platinum‐based derivative used to treat advanced colorectal cancer. Within the patient population on oxaliplatin therapy, a lower incidence of hematological adverse effects and gastrointestinal toxicity is noted, but severe neuropathic pain episodes characterized by increased cold and tactile hypersensitivity are present in ~95% of patients. This drug is also used to induce a rodent model of chemotherapy‐induced peripheral neuropathy (CIPN)‐related...

All‐Fullerene Electron Donor–Acceptor Conjugates

Published: 22 March 2019

Author(s): Marta Izquierdo, Benedikt Platzer, Anton J. Stasyuk, Olga A. Stasyuk, Alexander A. Voityuk, Sergio Cuesta, Miquel Solà, Dirk M. Guldi, Nazario Martín

Abstract The synthesis and characterization of a covalent all‐fullerene C60‐Lu3N@Ih‐C80 electron donor–acceptor conjugate has been realized by sequential 1,3‐dipolar cycloaddition reactions of azomethine ylides on Lu3N@Ih‐C80 and C60. To the best of our knowledge, this is the first time that two fullerenes behaving as both electron donor (Lu3N@Ih‐C80) and acceptor (C60) are forming an electroactive dumbbell. DFT calculations reveal up to 16 diastereomeric pairs, that is, 8 with syn and 8 with...

Fluorescent Triazole Urea Activity‐Based Probes for the Single‐Cell Phenotypic Characterization of Staphylococcus aureus

Published: 22 March 2019

Author(s): Linhai Chen, Laura J. Keller, Edward Cordasco, Matthew Bogyo, Christian S. Lentz

Abstract Phenotypically distinct cellular (sub)populations are clinically relevant for the virulence and antibiotic resistance of a bacterial pathogen, but functionally different cells are usually indistinguishable from each other. Herein, we introduce fluorescent activity‐based probes as chemical tools for the single‐cell phenotypic characterization of enzyme activity levels in Staphylococcus aureus. We screened a 1,2,3‐triazole urea library to identify selective inhibitors of...

A Stable, Non‐Corrosive Perfluorinated Pinacolatoborate Mg Electrolyte for Rechargeable Mg Batteries

Published: 22 March 2019

Author(s): Jian Luo, Yujing Bi, Liping Zhang, Xiaoyin Zhang, Tianbiao Leo Liu

Abstract Mg batteries are a promising energy storage system because of the physicochemical merits of Mg as an anode material. However, the lack of electrochemically and chemically stable Mg electrolytes impedes the development of Mg batteries. In this study, a newly designed chloride‐free Mg perfluorinated pinacolatoborate, Mg[B(O2C2(CF3)4)2]2 (abbreviated as Mg‐FPB), was synthesized by a convenient method from commercially available reagents and fully characterized. The Mg‐FPB electrolyte...

Polymer Dispersity Control by Organocatalyzed Living Radical Polymerization

Published: 22 March 2019

Author(s): Xu Liu, Chen‐Gang Wang, Atsushi Goto

Abstract Molecular weight distribution of polymers, termed dispersity (Đ), is a fundamental parameter for determining polymer material properties. This paper reports a novel approach for controlling Đ by exploiting a temperature‐selective radical generation in organocatalyzed living radical polymerization. The polymers with tailored Đ were synthesized in a batch system without the assistance of an external pump. A unique aspect of this approach is that Đ was tuneable from 1.11 to 1.50 in any...

Structural Transformation in Metal–Organic Frameworks for Reversible Binding of Oxygen

Published: 22 March 2019

Author(s): Jin‐Yue Zeng, Xiao‐Shuang Wang, Yong‐Dan Qi, Yun Yu, Xuan Zeng, Xian‐Zheng Zhang

Abstract Polycyclic aromatic derivatives can trap 1O2 to form endoperoxides (EPOs) for O2 storage and as sources of reactive oxygen species. However, these materials suffer from structural amorphism, which limit both practical applications and fundamental studies on their structural optimization for O2 capture and release. Metal–organic frameworks (MOFs) offer advantages in O2 binding, such as clear structure–performance relationships and precise controllability. Herein, we report the...

Development of Biocompatible and Antibacterial Collagen Hydrogels via Dialdehyde Polysaccharide Modification and Tetracycline Hydrochloride Loading

Published: 22 March 2019

Author(s): Xiaoyue Yu, Qijuan Yuan, Mingtao Yang, Ru Liu, Shichen Zhu, Jinling Li, Wanni Zhang, Juan You, Shanbai Xiong, Yang Hu

Abstract Nowadays, collagen hydrogels with both good physicochemical and antibacterial properties for tissue engineering have drawn broad attention. Herein, a biocompatible and antibacterial collagen hydrogel is developed via alginate dialdehyde (ADA) modification and tetracycline hydrochloride (TC) loading based on Schiff's base formation. Fourier transform infrared spectroscopy and X‐ray diffraction spectra suggest the maintenance of collagen structure integrity after ADA modification. The...

The Generation of Difluoroketenimine and Its Application in the Synthesis of α,α‐Difluoro‐β‐amino Amides

Published: 22 March 2019

Author(s): Rui Zhang, Zhikun Zhang, Qi Zhou, Lefei Yu, Jianbo Wang

Abstract Fluorine‐containing β‐amino acids and their derivatives have attracted significant attention due to their importance in life sciences. Herein the previously unknown difluoroketenimine, the analogue of the elusive difluoroketene, has been generated by the reaction of difluorocarbene and isocyanide, which further undergoes [2+2] cycloaddition with imine. The three‐component reaction affords α,α‐difluoro‐β‐amino amides in good yields. Mechanistic studies reveal the unique properties of...

Scalable Trehalose‐Functionalized Hydrogel Synthesis for High‐Temperature Protection of Enzymes

Published: 22 March 2019

Author(s): Priera H. Panescu, Jeong Hoon Ko, Heather D. Maynard

Abstract Herein, a scalable, two‐step synthesis of a trehalose hydrogel for the thermostabilization of enzymes is reported. A reaction between vinylbenzyl chloride and trehalose in base, followed by a redox‐initiated radical polymerization of the resulting mixture, produces the gel in 88% yield. The reaction scale can be increased 100‐fold while maintaining a 76% yield. Additionally, various solvents are investigated for purification, and more sustainable manufacturing solvents are selected....
