Photocatalytic Reductive Radical‐Polar Crossover for a Base‐Free Corey–Seebach Reaction

Published: 17 September 2020

Author(s): Karsten Donabauer, Kathiravan Murugesan, Urša Rozman, Stefano Crespi, Burkhard König

Abstract A metal‐free generation of carbanion nucleophiles is of prime importance in organic synthesis. Herein we report a photocatalytic approach to the Corey–Seebach reaction. The presented method operates under mild redox‐neutral and base‐free conditions giving the desired product with high functional group tolerance. The reaction is enabled by the combination of photo‐ and hydrogen atom transfer (HAT) catalysis. This catalytic merger allows a C−H to carbanion activation by the abstraction...

Stabilizing DNAzymes through Encapsulation in a Metal–Organic Framework

Published: 17 September 2020

Author(s): Huiye Zhong, Wei‐Shang Lo, Tiantian Man, Benjamin P. Williams, Dan Li, Sheng‐Yu Chen, Hao Pei, Li Li, Chia‐Kuang Tsung

Abstract DNAzymes are a promising class of bioinspired catalyst; however, their structural instability limits their potential. Herein, a method to stabilize DNAzymes by encapsulating them in a metal–organic framework (MOF) host is reported. This biomimetic mineralization process makes DNAzymes active under a wider range of conditions. The concept is demonstrated by encapsulating hemin‐G‐quadruplex (Hemin‐G4) into zeolitic imidazolate framework‐90 (ZIF‐90), which indeed increases the DNAzyme's...

Light‐Driven Active Ion Transport

Published: 17 September 2020

Author(s): Jinlei Yang, Pengchao Liu, Lianshan Li, Zhiyong Tang

Abstract Solar energy can be harvested by biological systems to regulate the directional transport of protons and ions across cells and organelles. Structural and functional bio‐mimic photo‐active ion nanofluidic conductors, usually in the forms of ion channels and ion pumps, have been increasingly applied to realize active ion transport. However, progress in attaining effective light‐driven active transport of ions (protons) has been constrained by the inherent limitations of membrane...

Oak‐Associated Negativicute Equipped with Ancestral Aromatic Polyketide Synthase Produces Antimycobacterial Dendrubins

Published: 17 September 2020

Author(s): Keishi Ishida, Gulimila Shabuer, Sebastian Schieferdecker, Sacha J. Pidot, Timothy P. Stinear, Uwe Knuepfer, Michael Cyrulies, Christian Hertweck

Abstract Anaerobic bacteria have only recently been recognized as a source of antibiotics; yet, the metabolic potential of Negativicutes (Gram‐negative staining Firmicutes) such as the oak‐associated Dendrosporobacter quercicolus has remained unknown. Genome mining of D. quercicolus and phylogenetic analyses revealed a gene cluster for a type II polyketide synthase (PKS) complex that belongs to the most ancestral enzyme systems of this type. Metabolic profiling, NMR analyses, and...

In Silico Anticipation of Metabolic Pathways Extended to Organic Chemistry Reactions: A Case Study with Caffeine Alkaline Hydrolysis and The Origin of Camellimidazoles

Published: 17 September 2020

Author(s): Victor Turpin, Mehdi A. Beniddir, Grégory Genta‐Jouve, Adam Skiredj, Jean‐François Gallard, Karine Leblanc, Pierre Le Pogam, Erwan Poupon

Abstract Camellimidazoles A–C were recently reported as natural substances in Keemun black tea. Although a “biosynthetic” route to these intriguing imidazole dimers was proposed from caffeine by the authors in this seminal report, we envisioned that a artefactual scenario, consisting of alkaline hydrolysis of caffeine and spontaneous cascade reactions with a methylene donor such as formaldehyde or methylene chloride, could also have led to their formation. To capture the diversity of molecules...
