DNA Encoded Libraries: A Visitor's Guide

Published: 17 January 2020

Author(s): Dillon T. Flood, Cian Kingston, Julien C. Vantourout, Philip E. Dawson, Phil S. Baran

Abstract In 1992, Brenner and Lerner hypothesized that individual chemical transformations could be encoded in DNA, allowing the rapid synthesis and screening of large collections of small molecules. Since their report, huge investments into the development of the DNA encoded library (DEL) technology have enabled the acceleration of the drug discovery process especially early phase discovery undertakings such as target validation and hit identification. As DEL lies at the nexus between...

Hollow Nano‐Mesosilica Spheres Containing Rhodium Nanoparticles Supported on Nitrogen‐Doped Carbon: An Efficient Catalyst for the Reduction of Nitroarenes under Mild Conditions

Published: 17 January 2020

Author(s): Shihan Wang, Jinyu Dai, Zhiqiang Shi, Zeshan Xiong, Zongtao Zhang, Shilun Qiu, Runwei Wang

Abstract Atom efficiency, low temperature, low pressure, and a nontoxic hydrogen source as a reducing agent are ideal reaction conditions for the reduction of nitroarenes. In this work, an efficient catalyst comprising hollow nano‐mesosilica spheres loaded with Rh nanoparticles supported on nitrogen‐doped carbon was developed. Rh nanoparticles were stabilized and uniformly dispersed by nitrogen atoms, and the inner N‐doped carbon shell was used to adsorb reaction substrates and improve...

Framework Effects on Activation and Functionalisation of Methane in Zinc‐Exchanged Zeolites

Published: 17 January 2020

Author(s): Meera A. Shah, Samuel Raynes, David C. Apperley, Russell A. Taylor

Abstract The first selective oxidation of methane to methanol is reported herein for zinc‐exchanged MOR (Zn/MOR). Under identical conditions, Zn/FER and Zn/ZSM‐5 both form zinc formate and methanol. Selective methane activation to form [Zn‐CH3]+ species was confirmed by 13C MAS NMR spectroscopy for all three frameworks. The percentage of active zinc sites, measured through quantitative NMR spectroscopy studies, varied with the zeolite framework and was found to be ZSM‐5 (5.7 %), MOR (1.2 %)...

Chemoenzymatic Synthesis of Sertraline

Published: 17 January 2020

Author(s): Lisa Marx, Nicolás Ríos‐Lombardía, Philipp Süss, Matthias Höhne, Francisco Morís, Javier González‐Sabín, Per Berglund

A chemoenzymatic approach has been developed for the preparation of sertraline, an established anti‐depressant drug. Ketoreductases (KREDs) were employed to yield a key chiral precursor. The bioreduction of the racemic tetralone exhibited excellent enantioselectivity ( 99 % ee) and diastereomeric ratio (99:1) at 29 % conversion (the maximum theoretical yield is 50 %) after 7 hours. The resulting (S,S)‐alcohol was efficiently oxidized to an enantiopure (S)‐ketone, an immediate precursor of...

Domino Hydroalkoxylation‐[4+2]‐Cycloaddition for Stereoselective Synthesis of 1,4‐Heterocycle‐Fused Chromenes: Rapid Access to the [6‐6‐7‐6] Tetracyclic Core of Cytorhizhins B–D

Published: 17 January 2020

Author(s): Santosh J. Gharpure, Santosh K. Nanda, Dipak J. Fartade, Dharmendra S. Vishwakarma

A substrate dependent regio‐ and stereoselective domino hydroalkoxylation‐formal‐[4+2] cycloaddition is described for the facile synthesis of linear as well as spirocyclic 1,4‐heterocycle‐fused chromene ketals. Enantiospecific synthesis of oxazepino chromene derivatives was successfully carried out using chiral pool amino alkynols. The developed hydroalkoxylation cascade offered rapid access to the spirocyclic [6‐6‐7‐6] tetracyclic core of cytorhizhins B–D with correct relative configuration.

Inhibition of allergen‐induced dermal eosinophilia by an oxoeicosanoid receptor antagonist in non‐human primates

Published: 17 January 2020

Author(s): Lisa A. Miller, Chantal Cossette, Shishir Chourey, Qiuji Ye, Chintam Nagendra Reddy, Joshua Rokach, William S. Powell

Background and Purpose 5‐Oxo‐6,8,11,14‐eicosatetraenoic acid (5‐oxo‐ETE), acting via the OXE receptor, is unique among 5‐lipoxygenase products in its ability to directly induce human eosinophil migration, suggesting its involvement in eosinophilic diseases. To address this hypothesis, we synthesized selective indole‐based OXE receptor antagonists. Because rodents lack an OXE receptor orthologue, we sought to determine whether these antagonists could attenuate allergen‐induced skin eosinophilia...

Photolysis of a hexane solution containing 1,1′‐ bis (trimethylsilylethynyl)ferrocene (1) and Fe (CO)5, under argon at 0 °C led to the formation of dinuclear complexes [Fe (CO)2{η2:η2 – C (SiMe3) = C(C5H4)FeC(C5H4) = C (SiMe3)Fe (CO)3}–μ–CO] (2) and [Fe (CO)2{η2:η2–C (SiMe3) = C(C5H5)–C(C5H5) = C (SiMe3)Fe (CO)3}–μ–CO] (3). DFT calculations support the experimentally observed demetalation of ferrocene unit of 2 to 3 in presence of water. These compounds were comprehensively characterized by IR...
